SNAP-8 (Acetyl Octapeptide-3) is a synthetic peptide designed as an extended version of Argireline (Acetyl Hexapeptide-8). It acts by modulating neurotransmitter release at the neuromuscular junction, thereby reducing muscle contraction–induced wrinkle formation, and is commonly studied in cosmetic applications as a topical alternative to botulinum toxin.

  • REFERENCES

SNAP-8 (Acetyl Octapeptide-3) is a synthetic cosmetic peptide derived from Argireline (Acetyl Hexapeptide-8), engineered to mimic the N-terminal end of SNAP-25, a substrate of the SNARE complex. By competing with natural SNAP-25, SNAP-8 reduces catecholamine-mediated neurotransmitter release at the neuromuscular junction. This mechanism leads to attenuated muscle contraction, thereby reducing the appearance of dynamic wrinkles and expression lines.

Structure

  • Type: Synthetic octapeptide
  • Sequence: Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp-NH₂
  • Molecular Weight: ~888 Da
  • Mechanism:

Competes with endogenous SNAP-25 for SNARE complex binding

Decreases synaptic vesicle release of acetylcholine

Reduces facial muscle contraction → smoother skin surface

Research

Cosmetic Applications

  • Demonstrated wrinkle depth reduction of up to 63% in 28 days (in topical studies).
  • Used in anti-aging creams, serums, and cosmetic formulations as a needle-free alternative to botulinum toxin.

Mechanistic Insights

  • Functions as a SNARE complex modulator.
  • Inhibits vesicle fusion and neurotransmitter release, similar to Argireline but with enhanced potency.

Safety Profile

  • Considered non-toxic and non-invasive.
  • Well tolerated in cosmetic applications with minimal adverse effects.
  • Effects are reversible upon discontinuation.

Summary

SNAP-8 is a synthetic octapeptide anti-wrinkle agent that:

  • Inhibits SNARE complex activity to reduce neurotransmitter release
  • Decreases muscle contraction, softening facial expression lines
  • Used widely in cosmetic dermatology as a topical alternative to Botox®

This peptide is supplied as a lyophilized powder. Store vials at 2–8 °C, protected from light and moisture. For long-term storage, keep unopened vials at −20 °C. After reconstitution, prepare under sterile conditions, refrigerate at 2–8 °C, and use promptly. Avoid repeated freeze–thaw cycles.
